As long as you do not cancel orders often there will be no downside in canceling an order. What is important is you cancel an order quickly so it does not become late to the end customer. If you cancel an order late your account could be flagged for causing a late order.

If your printer is waiting on a part to be shipped half way across the world or simply acting up with no end in sight. This would be a good time to cancel an order so the customer is able to get their order on time.
If someone wants a blue print and you only have 10 grams of blue filamnet left it would make sense at this point to cancel the order. Unless you are able to get the filament in time to still deliver to the customer in time.
Some 3D models are taken from online sources that are just not possible to print. The file may be corrupted or missing layers. If this is the case it is perfectly acceptable to cancel the order. Note if a print is just harder than normal and you do this often then your account will be flagged.
If you have 4 orders waiting to be worked but you leave to the Bahamas tomorrow. It is best practice to cancel the order. It is important to prioritize the customer getting the order on time.
